Project: Snow Leopard Genetics and Phylogeography

Techniques Learnt: Extraction of Snow Leopard DNA from non-invasively collected scats from the field and zoos, standardizing designed primers and using them in multiplex PCR. Sequence data troubleshooting and analysis.

Project: Standardisation of genetic markers for Snow Leopard (Uncia uncia )

I joined the multinational conservation genetics project on the charismatic and yet endangered big cat, Snow Leopard Uncia uncia. My job particularly was to extract DNA from the field collected faeces, amplifying neutral mitochondrial DNA markers through polymerase chain reactions (PCRs) and finally Sanger sequencing the amplified product. I edited the DNA sequences myself and then tried to understand their phylogenetics and phylogeography by reconstructing phylogenetic trees.
